| Working Principle |
A small drop of prepared boar semen is placed on a clean microscope slide. The cover glass is then carefully lowered onto the sample, preferably from one edge, to reduce the formation of air bubbles. Capillary action spreads the liquid semen beneath the cover glass and forms a thin observation layer. The microscope light passes through the slide, semen sample and cover glass, allowing sperm cells to be viewed through the objective lens. A properly prepared sample layer helps the operator observe sperm motility and distribution across the microscopic field. For motility evaluation, the slide and semen sample should be maintained at the temperature specified by the laboratory's semen-analysis procedure. The cover glass also separates the liquid specimen from the microscope objective, helping reduce accidental contamination of the optical system. |
